Appropriate sample handling is critical to successful ELISA measurements. Collection timing, manipulation procedures and storage conditions all directly shape final assay results. 1. Sputum samples
1.Weigh the viscous portion of collected sputum. Add 0.1 % dithiothreitol (DTT) solution at twice the volume of sputum to dissolve mucus. Repeatedly pipette the mixture, vortex for 15 seconds, then shake in a 37 °C water bath for 5 minutes.
2.Add PBS buffer equal to twice the original sputum volume and continue shaking for 15-20 minutes. Filter through a 150-mesh metal sieve, centrifuge at 1500 rpm for 10 minutes, and collect supernatant for testing.

2. Faecal samples
Collect relatively solid faecal material; overly watery samples are difficult to process and may compromise measurement accuracy. Obtain more than 50 mg of faecal specimen. Wash the sample three times with PBS, following a final mass-to-volume ratio of faeces to PBS of 1:9. Perform sonication (or mechanical homogenisation), then centrifuge at 5000 × g for 10 minutes and retain supernatant for detection.

3. Saliva samples
Place collected saliva inside centrifuge tubes and freeze at −70 °C for one hour. Thaw samples on ice, centrifuge at 2000 × g for 10 minutes and collect supernatant for testing.

4. Seminal plasma samples
Collect semen in sterile containers. Fresh semen normally appears thick and gelatinous. Allow complete liquefaction at room temperature or inside a 37 °C water-bath, driven by endogenous prostate fibrinolytic enzymes. Once fully liquefied, centrifuge semen at 4000 rpm for 10 minutes to separate seminal plasma for subsequent testing.

5. Skin tissue samples
Homogenise 20-50 mg skin tissue with pre-chilled PBS using a tissue homogeniser for full lysis. Centrifuge homogenate at 10 000 g for 20 minutes and collect supernatant for ELISA measurement.

6. Milk samples
Centrifuge milk at 12 000 g for 30 minutes under 4 °C conditions. Discard upper-layer fat and bottom-layer sediment, retain the middle liquid fraction for testing.

7. Cerebrospinal fluid and broncho-alveolar lavage fluid
After specimen collection, centrifuge at 1000 × g for 20 minutes. Harvest supernatant and store samples at −20 °C. Repeated freeze-thaw cycles must be avoided.

General sample-storage key notes
Sample processing aims to preserve target proteins. Since proteins are prone to denaturation and degradation, all manipulation steps should be carried out as gently as possible. Post-processing storage strongly impacts data quality:
Aliquot and seal processed samples to prevent repeated freeze-thaw events.
Storage duration limits: less than 1 week at 4 °C; maximum 1 month at −20 °C; maximum 2 months at −80 °C.
Allow specimens to slowly equilibrate to room-temperature before use; never rapidly thaw samples via heating, which causes protein inactivation and analyte breakdown.
